A Blood Group Test is used to determine a person’s blood type based on the ABO and Rh (Rhesus) systems. This test is essential for blood transfusions, organ transplants, pregnancy care, and medical emergencies.
Why is the Blood Group Test Done?
- To identify a person’s blood type (A, B, AB, or O).
- To check the Rh factor (positive or negative), which is important in pregnancy and transfusions.
- To ensure safe blood transfusions and prevent transfusion reactions.
- To assess Rh incompatibility during pregnancy, which can lead to hemolytic disease of the newborn (HDN).
Test Procedure

Blood Sample Collection
- A blood sample is drawn from a vein (usually in the arm) or a finger prick.

ABO Blood Grouping
- The blood is mixed with anti-A and anti-B antibodies to observe clotting (agglutination), determining the blood type:
- Type A → Clumps with anti-A serum.
- Type B → Clumps with anti-B serum.
- Type AB → Clumps with both anti-A and anti-B serums.
- Type O → No clumping with either serum.

Rh Factor Testing
- The blood is mixed with anti-Rh serum to check for Rh factor:
- Rh-positive (+) → Clumping occurs.
- Rh-negative (-) → No clumping.
Interpreting the Results
The test determines the blood type in one of the following combinations:
- A+ (A positive) or A- (A negative)
- B+ (B positive) or B- (B negative)
- AB+ (AB positive) or AB- (AB negative)
- O+ (O positive) or O- (O negative)
